The serum levels of carboxylated and undercarboxylated osteocalcin in children with cystic fibrosis

Ambroszkiewicz Jadwiga , Sands Dorota , Gajewska Joanna , Chelchowska Magdalena , Laskowska-Klita Teresa

Introduction: Osteocalcin (OC) is the noncollagenous protein of bone matrix produced by osteoblasts which play an important role in bone metabolism. In its carboxylated form (c-OC) osteocalcin binds to hydroxyapatite in bone and plays a regulatory role in bone formation and mineralization. In contrast, undercarboxylated OC (uc-OC) binds less effectively to hydroxyapatite and a significant association has been found between fracture incidence and uc-OC in elderly subjects. Unde...

Introduction: Obesity is the direct cause of a number of immediate problems during childhood. Among them the high prevalence of fractures and joint problems in obese children was observed. Leptin may influence the development and bone remodeling in children, but this phenomenon is not well understood. Therefore, we studied the relationships between leptin and bone turnover markers, growth factors and anthropometric parameters in obese children.Descriptio...

Recurrent fractures and low bone mass in a patient with new mutation of LRP5 gene

Rusinska Agnieszka , Borowiec Maciej , Mlynarski Wojciech , Antosik Karolina , Michalus Izabela , Golec Joanna , Chlebna-Sokol Danuta

In recent years, the important role in bone remodelling Wnt/β-catenin pathway is highlighted. Key receptor of this pathway is LDL receptor-related protein 5 (LRP5). It was demonstrated in adults that polymorphism in LRP5 gene was associated with bone mineral density and fracture risk. So far no such studies were conducted in children. The aim of the study was the analysis of LRP5 and COL1A1 genes in a patient with r...

Osteogenesis imperfecta: diagnostic difficulties due to clinical symptoms diversity: the results of own studies

Rusinska Agnieszka , Jakubowska-Pietkiewicz Elzbieta , Michalus Izabela , Kurnatowska Olga , Rychlowska Ewa , Golec Joanna , Chlebna-Sokol Danuta

Osteogenesis imperfecta (OI) is a genetic bone dysplasia characterized by recurrent fractures and reduced bone mineral density. The severity of its symptoms varied from very mild to severe, which strongly affect the quality of life and cause premature death. The aim of the study is to compare the clinical symptoms of different types of osteogenesis imperfecta and to present diagnostic difficulties based on the analysis of our patients.</p...

The significance of Bcl-2-associated athanogene-1 (BAG-1)-mediated protein interactions in osteoblast development

Greenhough Joanna , Papadakis Emmanouil , Cutress Ramsey , Townsend Paul , Oreffo Richard , Tare Rahul

BAG-1 interacts with diverse proteins including heat shock proteins (HSC70/ HSP70) and nuclear hormone receptors to modulate cell proliferation, differentiation and apoptosis. We have shown that bone marrow stromal cells (BMSCs) of Bag-1+/− female mice display reduced BMP-2-directed osteogenic differentiation potential. Estradiol binds to estrogen receptors (ERs) and is functionally involved in BMP-directed osteogenic differentiation of osteoprogenito...

The assessment of bone regulatory pathways in children with malignant bone tumors

Ambroszkiewicz Jadwiga , Gajewska Joanna , Chelchowska Magdalena , Rogowska Elzbieta , Rychlowska-Pruszynska Magdalena

Objectives: As the receptor activator of the nuclear factor κB (RANK)/receptor activator of the nuclear factor κB ligand (RANKL)/osteoprotegerin (OPG) cytokine system is essential for osteoclastogenesis and Wnt signaling pathway for osteoblastogenesis, we decided to assess these two main bone regulatory pathways in patients with malignant bone tumors on the completion of anticancer therapy. Methods: The study included 35 patients (median age 15....

Effect of paracetamol on bone mechanical properties in rats

Cegiela Urszula , Janas Aleksandra , Folwarczna Joanna , Pytlik Maria , Sliwinski Leszek , Koziel Angelika , Koziol Marzena , Ludwig Rafal

Results of epidemiological studies indicate that paracetamol use may be a risk factor for fracture. The exact mechanism of paracetamol action remains unclear, but it is known to inhibit COX-2. Orchidectomy-induced androgen deficiency was reported to increase COX-2 expression and prostanoid release in rats. The aim of the present study was to investigate the effect of paracetamol on bone mechanical properties in rats with normal androgen levels and with androgen deficiency indu...

Doxazosin prevents the development of estrogen deficiency-induced osteoporosis in rats

Pytlik Maria , Janas Aleksandra , Folwarczna Joanna , Cegiela Urszula , Sliwinski Leszek , Konieczna Agnieszka , Szklanny Ewa , Adamski Lukasz

In postmenopausal women, estrogen deficiency not only interferes with the metabolism of the bone tissue, but also adversely affects the cardiovascular system and can cause hypertension. Doxazosin is often used in the treatment of resistant hypertension, especially in patients with renal failure and hyperlipidemia. Vasodilator effect of doxazosin is associated with selective blocking of α1 receptors in the smooth muscle cells of resistance arterioles. The results of epidem...

Mast cells regulate inflammation and bone regeneration in fracture healing

Kroner Jochen , Kovtun Anna , Messmann Joanna , Strauss Gudrun , Seitz Sebastian , Schinke Thorsten , Dudeck Anne , Ignatius Anita

Mast cells (MCs) are pro-inflammatory sensor and effector cells of the immune system. MCs seem to play a role in bone metabolism, because patients with systemic mastocytosis develop osteoporosis. MCs are present in the fracture callus during bone healing, however, their function has not yet been investigated. Here, we examined the role of MCs in the inflammatory and repair phase during fracture healing.Male 12-week old MC deficient mice (Mcpt5-Cre R-DTA)...
