Denosumab's dynamic CTX profile is maintained over 6 years of treatment: first 3 years of the FREEDOM extension study

Roux C , McClung MR , Franchimont N , Adami S , Ebeling PR , Reid IR , Resch H , Weryha G , Daizadeh N , Wang A , Wagman RB , Eastell R

Denosumab (DMAb) has a unique profile of bone resorption inhibition: CTX decreases rapidly by 3 days and inhibition is released at the end of the 6-month dosing interval, when DMAb serum levels decrease (McClung NEJM 2006). The dynamic CTX inhibition profile is not curtailed by continued treatment. In the 3-year FREEDOM study, CTX values at 6 months were influenced by baseline CTX values and days since the 1st injection (Eastell JBMR 2011). With 3 additional ...

Resolution of effects on bone turnover markers and bone mineral density after discontinuation of long-term bisphosphonate use

Benhamou Claude , De Villiers Tobias , Johnston C Conrad , Langdahl Bente , Saag Kenneth , Denker Andrew , Pong Annpey , McGinnis John P , Rosenberg Elizabeth , Santora Arthur

Relatively little is known about immediate consequences of continuing vs interrupting long-term bisphosphonate treatment. This report describes changes in bone turnover and BMD in a 1-year, dose-finding trial of the calcium-sensing receptor antagonist MK-5442 in postmenopausal, BP-treated women, randomized to continued alendronate 70 mg weekly, switch to placebo, or switch to MK-5442. Recruited women (n=526) had taken alendronate for ≥12 months and an oral BP fo...

Transdermal delivery of BA058, a novel analog of hPTHrP (1-34), with a short wear time patch in preclinical and clinical studies

Hattersley Gary , Hansen Kris , Determan Amy , Brown Ken , Mckay Kate , Guerriero Jonathan , McCarthy Dan , Lyttle C Richard , St L O'Dea Louis

BA058 is being developed as an anabolic therapy for the treatment of osteoporosis. Daily BA058 SC injection has produced promising safety and efficacy results in early clinical studies, and is currently enrolling in a Phase 3 fracture prevention study. There is, however, a significant need for an alternative to injection that improves patient convenience and compliance. We have investigated the use of a solid Microstructured Transdermal System (3M) for transdermal (TD) deliver...

Influence of nutritional status in the bone mass assessed by phalangeal quantitative ultrasound of girls aged 7–10 years old

Bertapelli Fabio , Goncalves Ezequiel M , Barbeta Vinicius J O , Krahenbuhl Tathyane , Ramalho Luis C B , Martin Juan E Samur-San , Ribeiro Roberto R , Guerra-Junior Gil

The childhood obesity is a global epidemic. Some studies with pre-pubertal children reported a positive relationship between fat mass and bone. However, there are no available data on the influence of the nutritional status on parameters of quantitative ultrasound (QUS). The aim of this study was to evaluate the influence of nutritional status in bone mass assessed by QUS of proximal phalanges in girls. The bone mass parameter, amplitude dependent speed sound (AD-SoS) in meter...

An unusual presentation of progressive osseous heteroplasia in a 7-year-old female child

Schrander D E , Welting T J , Schrander J J P , van Rhijn L W , Korver-Keularts I , Schrander-Stumpel C T R M

Background: Progressive osseous heteroplasia (POH) (OMIM 166350) is a rare autosomal dominant condition, characterized by heterotopic ossification of the skin, subcutaneous fat and deep connective tissue. This condition is distinct from Albright’s hereditary osteodystrophy or Mccune–Albright syndrome (AHO) (OMIM 103580) and fibrodysplasia ossificans progressiva (FOP) (OMIM 135100).Presenting problem: We present an unu...

WNT5A has anti-prostate cancer effects and protects against bone metastases

Thiele Stefanie , Gobel Andy , Hippauf Sandra , Rachner Tilman D. , Muders Michael , Fussel Susanne , Bernhardt Ricardo , Jakob Franz , Rauner Martina , Hofbauer Lorenz C.

Prostate cancer (PCa) is the most common cancer type in older men and often metastasizes to bone in advanced stages. Wnt proteins are implicated in carcinogenesis and especially WNT5A has been discussed to influence the clinical outcome of various cancer types, including PCa. In addition, WNT5A stimulates osteogenic differentiation and may thus not only be involved in PCa development, but also in the formation of subsequent skeletal metastasis. Here, we determined the role of ...

Abnormal trabecular micro-architecture and bone mechanical properties in adolescent idiopathic scoliosis: a case–control study with high-resolution peripheral quantitative computed tomography and finite element analysis

Cheuk Ka Yee , Chen Huanxiong , Yu Fiona , Lam Tsz Ping , Lee Simon K M , Ng Bobby K W , Cheng Jack C Y

Adolescent idiopathic scoliosis (AIS) is associated with low bone mineral density (BMD) which has been reported to be an important prognostic factor for curve progression. In addition to BMD, high-resolution peripheral quantitative computed tomography (HR-pQCT) allows measurement of volumetric BMD, bone morphology, trabecular micro-architecture, and evaluation with structure model index (SMI) and finite element analysis (FEA) for mechanical property assessment. The aim of this...

Lack of PEDF within the bone matrix is associated with osteoidosis and abnormally high bone mineral content

Fratzl-Zelman Nadja , Schmidt Ingo , Roschger Paul , Roschger Andreas , Reich Adi , Glorieux Francis H. , Wagermaier Wolfgang , Fratzl Peter , Klaushofer Klaus , Marini Joan C. , Rauch Frank

Background and methods: Pigment epithelium-derived factor (PEDF) is a potent antiangiogenic factor, ubiquitously expressed and secreted in human tissues. Hypertrophic cartilage and osteoblasts express PEDF that binds to type I collagen and glycosaminoglycans in the extracellular matrix. Two rare forms of osteogenesis imperfecta (OI) with intact collagen synthesis are associated with PEDF deficiency. Histological observations revealed excessive osteoid formation and prolonged m...

Cheuk Ka Yee , Cheung Franco , Lee WayneY W , Hung Vivian , Tam Elisa , Lam Tsz Ping , Ng Bobby K W , Lee Simon K M , Cheng Jack C Y

Objectives: This study aimed to evaluate detailed bone profiles during puberty for Chinese adolescents in Hong Kong using high-resolution peripheral quantitative computed tomography (HR-pQCT) and dual-energy x-ray absorptiometry (DXA).Methods: 333 healthy girls and 231 healthy boys (12–16-years-old) were recruited from secondary schools. Areal bone mineral density (aBMD) of bilateral femoral necks were measured by DXA while bone quality parameters w...
