A Danish database initiative was established in 2003 with the purpose of evaluating patients treated with teriparatide. After the introduction in 2006 of 1–84-PTH, these patients were also included. A total of 1494 patients from 19 centers were reported to the database, until closure in 2011. The reported patients correspond to a third of all patients in Denmark receiving anabolic treatment during this time period. With regard to age and sex the reported patients do not d...

Cytokine levels in the bone marrow are not just a reflection of the levels in serum: suggesting that bone marrow constitutes an independent compartment

Harslof Torben , Sorensen Lotte , Pedersen Steen , Langdahl Bente

Bone marrow is composed of cells developed within the marrow compartment as well as blood supplied from the general circulation. Bone tissue is affected by cytokines within the bone marrow. We investigated if the levels of different bone acting cytokines differ between bone marrow and blood from the peripheral circulation.Fifty-three healthy postmenopausal women participated in a study designed to evaluate the effect of rosiglitazon...

A polymorphism in the TGF-β1 gene affects TGF-β1 secretion

Husted Lise , Sorensen Lotte , Stenkjaer Liselotte , Langdahl Bente

Transforming growth factor (TGF)-β1 is the most abundant growth factor in human bone. Several polymorphisms have been described in the TGF-β1 gene (TGFB1). We have previously shown that individuals with the CC genotype of the T29C polymorphism have higher bone mass at the femoral neck. The T29C polymorphism causes a change from leucine to proline at codon 10, which is located in the hydrophobic α-helical part of the signal peptide....

Is recovery of lactation-induced bone loss influenced by immobilization in mice?

Givskov Sorensen Thomas , Bruel Annemarie , Skovhus Thomsen Jesper

Lactation in both humans and mice is associated with a substantial bone loss, in mice this bone loss is recovered within 4 weeks after weaning. This recovery is considered to be the most rapid physiological bone formation in adult life. The aim of the study was to investigate the effect of the post-lactational anabolic response on a disuse bone loss induced by Botulinum toxin (BTX).Forty-eight NMRI mice were divided into the following groups: pregnan...

Reference curves for bone health index for Han children from five large cities in China, and a comparison to Asian-American children

Thodberg Hans Henrik , Zhang Shao-Yan

Objective: The bone health index (BHI) has previously been shown to be 1.5% lower in Asian children than in Caucasian children, both living in USA. The aim of this study is to present reference curves for BHI of Chinese Han children and to compare to Asian children living in USA.Method: BHI is derived from the cortical thickness in the three middle metacarpals. It is determined with the BoneXpert medical device, which automatically analyses a standard bo...

Bone health index: Swiss children have less in the bank than a generation ago

Thodberg Hans Henrik , Martin David D , Caflisch Jon , Jenni Oskar

Objective: The aim of this study is to compare the bone health index (BHI) for healthy Swiss children born in 1955 with healthy Swiss children born a generation later.Method: BHI is derived from the cortical thickness in the three middle metacarpals. It is determined with the BoneXpert medical device, which automatically analyses a standard bone age hand radiograph. The measurement result is independent of the sharpness of the image. The image data are f...

Reference values of cortical thickness, bone width, and Bone Health Index in metacarpals of children from age 0 y, as determined with an extension of the fully automated BoneXpert bone age method

Thodberg Hans Henrik , Thrane Peter , Martin David D.

Objective: The BoneXpert method for automated bone age determination from hand X-rays also determines the cortical thickness T and the bone width W in the three middle metacarpals. From these, the method derives the cortical area A=π W T (1 – T/W), the metacarpal index MCI =A/(WW) and the Bone Health Index. Recently, the method has been extended down to new-borns, and the aim of this study is to report reference curves for these bone measures.M...

Low serum thyrotropin level and duration of suppression as a predictor of major osteoporotic fractures – the openthyro register cohort

Abrahamsen Bo , Jorgensen Henrik L , Laulund Anne S , Nybo Mads , Brix Thomas H , Hegedus Laszlo

Objective: To assess the relationship between thyrotoxicosis and osteoporotic fractures in men and women.Design: Register-based cohort study in patients with a TSH measurement in the region of Funen 1996–2010. All determinations were done in the same lab serving all hospitals and GP practices. Persons with raised TSH or a history of thyroid/pituitary disease were not included. Results: The study population consisted of 222 138 ...

Regulation and function of lentiviral-mediated TCIRG1 expression in osteoclasts from infantile malignant osteopetrosis patients

Lofvall Henrik , Thudium Christian Schneider , Moscatelli Ilana , Montano Carmen , Schulz Ansgar , Richter Johan , Henriksen Kim

Infantile malignant osteopetrosis (IMO) is a rare, lethal, recessive disorder characterized by dysfunctional osteoclasts. TCIRG1, encoding the osteoclast V-ATPase, is mutated in 50% of IMO patients. We have previously shown that the resorptive function in osteoclasts derived from IMO patients can be restored in vitro by expressing TCIRG1 using a lentiviral vector. In this study, we aim to investigate the cellular response to vector-derived TCIRG1 expression and to det...

Vitamin D from the marine inuit diet and markers of inflammation – a population-based survey in Greenland

Schaebel Louise , Bonefeld-Jorgensen Eva , Laurberg Peter , Vestergaard Henrik , Andersen Stig

Background: The traditional Inuit diet in Greenland consists mainly of fish and marine mammals. The marine diet is an important source for vitamin D in Greenland. Vitamin D has anti-inflammatory capacity but markers of inflammation were high in Inuit with a high intake of marine food items. Yet, the effect of vitamin D on inflammation in Inuit remains unsettled. Objectives: To investigate the association between vitamin D and markers of inflammation in a ...
